Our lab is interested in the conformational dynamics of multi-subunit ATPase complexes of the AAA+ (ATPases associated with various cellular activities) superfamily. We aim to obtain a structural view of these molecular machines by combining single particle cryo electron (EM) microscopy with biochemical and biophysical analysis.
The aim of the project is to solve the structures of ClpV and ClpC (Hsp100) oligomers at the highest possible resolution using cryo EM. By recording structural snapshots of their movements we want to understand the working mechanism of the oligomer. Focus in this project will be to optimize current data collection and image processing steps to achieve high resolution cryo EM structures.
